Lens Assays on Diabetic and Galactosemic Rats Receiving Diets That Modify Cataract Development.

چکیده
Cataract formation is delayed or prevented in diabetes and galactosemia by diets rich in fat and protein. This is accomplished in the presence of high levels of blood sugar and lens sugar alcohols. Diabetic rats on a high-fat diet for 235 days had lenses that were clear to the naked eye, whereas control animals developed cataracts in a median time of 65 days. The lens protein level of treated animals was about 90 per cent of normal. There was no evidence of lens swelling; however, the concentration of lens amino acids ivas low.

Galactose-induced retinal microangiopathy in rats.
PURPOSE The suitability of the galactose-fed rat as a model of diabetic retinopathy was examined in nondiabetic rats fed diets enriched with either 30% or 50% galactose for up to 2 years. METHODS Retinal capillaries were examined by light and electron microscopy, and the prevalence or severity of diabetic-like lesions was quantitated. RESULTS Histologic evaluation of trypsin digests of reti...
متن کاملDiosgenin, a Novel Aldose Reductase Inhibitor, Attenuates the Galactosemic Cataract in Rats
OBJECTIVE To seek efficient aldose reductase inhibitors (ARIs) with excellent in vitro and in vivo biological activities against rat galactosemic cataract. METHODS The method was firstly optimized to screen strong ARIs from nonoriented synthetic compounds and natural extracts. Then, diosgenin was assessed on osmotic expansion of primarily cultured lens epithelial cells (LECs) induced by galac...
متن کاملA simple and stable galactosemic cataract model for rats.
Rat galactosemic cataract is commonly used in the investigation of sugar cataract. In current study, 21-day sprague-dawley (SD) rats were randomly divided into two groups (n=42), which were fed by normal water and galactose solution (12.5%-10%) for 18 days respectively. Every 3 days, lens opacity was observed by a slit lamp, and 6 rats of each group were executed for the analysis of aldose redu...
متن کاملPotential of Allylmercaptocaptopril as an Anti Cataract Agent against Galactosemic Cataract in Rats
Aim: Allylmercaptocaptopril (AMC) is a covalently bonded product of allicin and captopril has been evaluated for its anticataract activity against selenite induced cataract in experimental animals. We wanted to evaluate its anticataract potential in galactosemic cataract to elucidate biochemical mechanism to appraise its activity. Method: We examined the protective effect of AMC in both in vitr...
متن کاملThe Effect of Silybum marianum (L.) Gaertn. Seed Extract (Silymarin) on Galactose Induced Cataract Formation in Rats
Background: Increased oxygen free radical and reduced glutathione level in the eye lens are important risk factor for cataract formation. The antioxidative property and increasing cellular and extra cellular glutathione level have been reported by several herbal medicines including silymarin. Objective: In present interventional study Silybum marianum L. seed extract (silymarin) was tested a...
